Stain solutions
Acid Alcohol
Concentrated hydrochloric acid 3 ml
Ethanol (95%) 97 ml
Alberts Stain Solution:
Malachite green 0.2 gm;
Toluidine Blue 0.15 gm;
95% ethanol 2 ml;
Glacial acetic acid 1 ml;
Distilled water 100 ml.
Dissolve the dyes in the ethanol. Mix the acid with the water and add to the dye solution.
Let stand for 24 hours and then filter.
Ammoniacal silver nitrate
Silver nitrate 5 g
Distilled water 100 ml
Take 90 ml of the above solution and add strong ammonia solution drop by drop until the precipitate that forms just dissolves
Then add the rest of the silver nitrate solution drop by drop until the reagent remains slightly turbid even after shaking
Protect from the light
Stable for several weeks
Carbol Fuchsin
Solution A
Basic fuchsin 10 g
Ethanol (95%) 100 ml
Mix and dissolve and keep at 37 C overnight in a stoppered bottle
Solution B
Phenol 5 g
Distilled water 100 ml
Mix and dissolve
For use add 10 ml of solution A into 100 ml of solution B and mix
Crystal Violet Solution
Dienes Stain
Methylene blue 2.50 gm
Azure II 1.25 gm
Maltose 10.00 gm
Sodium carbonate 0.25 gm
Distilled water 100 ml
Kirkparticks Fixative
Absolute ethanol 60 ml
Chloroform 30 ml
Formaldehyde (40%) soln 10 ml
Loefflers Methylene Blue
Grams (Lugols) Iodine Solution
Plimmers Mordant
Iron Tannate Mordant
Giemsa Stain solution
Gram alcohol solution
Ethanol
water
Malachite Green Solution
Muirs Mordant
Neissers stain
For use, mix 20 ml of solution A and 10 ml of solution B
NeissersSolution A:
Methylene blue 0.1 gm;
95% ethanol 5 ml;
Glacial acetic acid 5 ml
Distilled water 100 ml.
Dissolve the dye in the water and add the acid and ethanol
Neissers Solution B:
Crystal violet 0.33 gm;
95% ethanol 3.3 ml
Distilled water 100 ml.
Dissolve the dye in the ethanol- water mixture
Safranin Solution
Safranin 0.5 g
Water 100 ml
Sudan Black B Solution
Sudan Black B 0.3 gm
95% ethanol 75 ml
Distilled water 25 ml
Dissolve the Sudan Black B in 95% ethanol then add water and mix thoroughly
