Principle | Gluconate can be converted to 2-ketogluconate which can be detected by the appearance of a reducing substance in the medium. The test is useful in identifying Pseudomonas aeruginosa, Enterobacter species and those Klebsiella with IMViC reaction --++. |

Method | Inoculate gluconate broth and incubate for 2 days. Add 1 ml of Benedict's solution, mix and boil for 10 min. Clintest tablets (Ames Co) can replace Benedict's solution |
Results | The formation of a brown, orange or tan preciptate is a positive reaction |
