Method | Two biologically equivalent guinea pigs are used; one of these (control animal) is given an intraperitoneal injection of 1000 units of diphtheria antitoxin 12 hours before inoculation. Both guinea pigs are partially depilated and 0.2 ml of a 12 hour culture of the organism in broth is injected intracutaneously into each animal. Virulent diphtheria microbes cause inflammation and swelling at the injection site within 24-48 hours. Within 3-4 days the inflammation subsides leaving a necrotic area which later forms a scab or eschar. There is no effect on the control animal. |
