Principle Some strains of Staphylococci produce agents called Staphylocoagulase that promote the clotting of plasma. Staphylocoagulase is made up of free and bound coagulase
Cautions Do not use plasma that contains EDTA

The type of plasma may affect the result. The citrate in citrated plasma may be removed by some citrate utilizing bacteria which may cause a clot to form after prolonged incubation.

Heparin can be added to citrated plasma to prevent this problem.

Production of large amounts of fibrinolysin wil dissolve the clot

Method . Slide methods detect just bound coagulase while tube methods detect both forms.
